Quantifying lung virus titer reduction following immunization. Lung tissue homogenates were collected and serially diluted in serum-free DMEM prior to plaque assay. Diluted virus mixtures were inoculated into confluent monolayers of HEp-2 cells to initiate virus infection. RSV rA2-line19F plaques were visualized using the DAB substrate. Plaques were counted and used to calculate the lung virus titer from each sample. Data are presented as mean ± SD from experiments performed in triplicate and statistical significance was determined by one-way ANOVA with Tukey’s multiple comparisons post hoc test (** p < 0.01, *** p < 0.001)
