Increased LPS-induced and/or nigercin-induced NLRP3 inflammasome-mediatedIL-1β release in peripheral blood mononuclear cells (PBMCs) from patients with asthma is not affected by sputum inflammatory phenotype. PBMCs from patients non-severe (NSA) and severe stable (SA(S)) asthma were A pre-cultured with media for 4 h before being stimulated with nigericin for 1 h, or B pre-stimulated with LPS for 4 h before stimulation with nigericin for 1 h, and the effects on IL-1β release were assessed in culture supernatants. The effects of nigericin or LPS + Nigericin stimulation on IL-1β release were assessed following stratification of subjects by sputum inflammatory phenotype. Data are presented as means ± SEM (N = 24–41)
