MyD88 in lung structural cells is important for regulating ODE-induced airway hyper-responsiveness (AHR). MyD88 bone marrow chimeric mice (donor → recipient) were treated with ODE (solid line) or saline (dotted line), and at three hr following i.n. inhalation treatment mice were tracheostomized, mechanically ventilated, and AHR to aerosolized methacholine (0, 3, 6, 12, 24, 48 mg) was measure and expressed as mean (±SEM) total lung resistance (RL). Chimeric groups depicted: a KO→WT; b WT→KO; c WT→WT; d KO→KO. Statistical difference between ODE and saline treatment groups was determined by two-way ANOVA (because there are 2 independent variables: treatment group and methacholine dose). If p-value less than 0.05, differences between saline and ODE at each methacholine doses were determined by t-test and asterisks denote significance (*p < 0.05, **p < 0.01, ***p < 0.001). N = 4–6 mice/group
