TRPV4 regulates stretch-induced release of IL-6. E17 epithelial cells were isolated and seeded on bioflex plates coated with fibronectin. 24 hours later, cells were exposed to 20 % cyclic stretch for 48 h in the presence or absence of the vehicle DMSO, the TRPV4 agonist GSK1016790A [100 nM] or TRPV4 antagonist HC-067047 [1 μM]. Unstretched cells served as controls. Supernatants were collected and processed to assess IL-6 concentrations by ELISA, as described in methods. Values are mean ± SEM from 5 different experiments. Results were normalized to the cell lysate protein concentrations. *p < 0.05 vs control vehicle; **p < 0.01 vs stretch vehicle. Tukey-Kramer Multiple Comparisons Test.
