Isolation and phenotypic characterization of mDC subsets. ( A ) PBMCs were gated on SSC and FSC to exclude dead cells and debris. Cells were then gated to exclude lineage positive cells and pDCs. Lineage (Lin)^-, BDCA-2^- cells were then gated for expression of BDCA-1 and BDCA-3. BDCA-1⁺ mDCs were sorted as Lin^-, BDCA-2^-, BDCA-1⁺, BDCA-3^dim and BDCA-3⁺ mDCs were sorted as Lin^-, BDCA-2^-, BDCA-1^-, BDCA-3^hi. ( B ) Post-sort purity for all sorted cells was ≥95%.
