EMMPRIN blocking antibody attenuates TGF-β1 induced lung fibroblast proliferation, migration and differentiation to myofibroblast: NHLF were stimulated with TGF-β1 (0-10 ng/ml) in the presence of either EMMPRIN functional blocking antibody or IgG control antibody examined for (a) Cells proliferation using [3H] thymidine incorporation; (b) Cell migration using a modified Boyden chamber assay. Each bar represents means ± SEM of 3 independent experiments, each run in duplicate, # p = 0.002 or ** p < 0.001 comparing EMMPRIN functional blocking antibody to IgG control antibody treated cells; and (c) Cell differentiation using Western blot for α-SMA of cell lysates from NHLF treated with TGF-β1 (5 and 10 ng/ml) in the presence of either EMMPRIN functional blocking antibody or IgG antibody. β-Tubulin was used as the loading control.
