The effect of IL-33 treatment on fibrotic marker expression by human PCLS. PCLS from normal adjacent (NAT) and IPF lung tissue were stimulated with 30 ng/ml of IL-33 or 2 ng/ml of TGFβ. FN1, ACTA2 and COL1A1 mRNA expression by NAT and IPF PCLS was measured by qPCR and normalised to the unstimulated media alone control for each individual donor (A). Fibronectin expression was assessed by western blot (10 μg total protein/lane) following SDS-PAGE analysis (B) and quantified relative to GAPDH via densitometry (C).
