Effects of Epac regulators on SOCE in ASMCs. Intracellular Ca2+ concentration was determined with confocal Ca2+ measurements, shown are representative time courses of calcium fluorescence changes. Thapsigargin (TpG) was used to induce the depletion of ES/SR Ca2+ store, and store-operated Ca2+ entry was induced by 2 mM CaCl2-containing bath solution. Ca2+ fluorescence changes in mouse ASMCs treated with 0, 1, 10, 100 μM 8pCPT or ESI-09 (a). Summarized data indicated that both 8pCPT and ESI-09 had no effect on TpG-induced Ca2+ release in Ca2+-free solution. 8pCPT inhibited store-operated calcium entry (SOCE) only at 100 μM, whereas ESI-09 could promote SOCE at 10 and 100 μM in ASMCs (b). 10 μM SKF-96365, the store-operated Ca2+ channel blocker, inhibited the promoting effects of ESI-09 on ASMCs proliferation after cultured for 48 h (c).
