Effect of P2X7R or P2Y2R silencing with siRNA on MUC5AC release in poly(I:C)-treated NCI-H292 cells. a, c Effect of P2X7R or P2Y2R silencing with siRNA in NCI-H292 cells was evaluated by immunoblotting for 24 h after transfection. The amount of P2X7R or P2Y2R was assessed by densitometry. b, d Effect of P2X7R or P2Y2R silencing with siRNA on the release of MUC5AC in poly(I:C)-treated cells. Cells were treated with non-targeting, P2X7R or P2Y2R siRNA 48 h prior to the treatment with 10 μg/ml poly(I:C) or vehicle. After 24 h, supernatants were harvested and assayed for MUC5AC release by ELISA. The data were normalized by the values to control siRNA at 0 μg/ml poly (I:C). The data are expressed as the mean ± SEM for three to four separate experiments.
