Effect of P2R antagonists on MUC5AC production and release in poly(I:C)-treated NCI-H292 cells. a-c Effect of a non-selective antagonist of P2R, suramin, on MUC5AC release in poly(I:C)-treated cells. Cells were treated with 10 μM suramin or vehicle 30 min prior to the treatment with 10 μg/ml poly(I:C). After 24 h, whole cells were harvested, and the expression of MUC5AC was assessed with Real-time PCR (a), and the intracellular protein of MUC5AC was assayed by ELISA (b). c Various concentrations of suramin were added 30 min before 10 μg/ml poly(I:C) treatment. After 24 h, the supernatants were harvested and assayed for MUC5AC release by ELISA. d, e Effect of P2X7R antagonists on MUC5AC release in poly(I:C)-treated cells. Various concentrations of a P2X7R antagonist, Brilliant Blue G (BBG), or a specific P2X7R antagonist, A438079, were added 30 min before 10 μg/ml poly(I:C) treatment.
