Desipramine and fluoxetine repress NF-ĪŗB and AP-1 activities. A, A549 cells were transfected with a NF-ĪŗB luciferase gene construct and pJ7-LacZ. After transfection, cells were either left untreated (UT) or pretreated or not for 1 h with desipramine (DMI, 10-5 M) or fluoxetine (Fluo, 10-5 M), and further stimulated for 4 h with TNF-Ī± (10 ng/ml). NF-ĪŗB activity was normalised and that induced by TNF-Ī± alone (47 Ā± 8 fold induction) was given the nominal value of 100 %. Data are shown as the percentage of activity relative to this nominal value. *p < 0.001 versus TNF-Ī± alone, by Welch t test. B, A549 cells were transfected with an AP-1 luciferase gene construct and pJ7-LacZ. After transfection, cells were either left untreated (UT) or pretreated or not for 1 h as in A, and further stimulated for 4 h with TPA (10 ng/ml). AP-1 activity was normalised and that induced by TPA alone (2.3 Ā± 0.1 fold induction) was given the nominal value of 100 %. Data are shown as the percentage of activity relative to this nominal value. *p < 0.01 versus TPA alone, by Welch t test.
