Loss of TRIP-1 is associated with increased phosphorylation of the pro-survival protein Akt, and increased Akt activity drives α-SMA expression in cells with low TRIP-1 expression. A: HLF-F were transfected with control siRNA or TRIP-1 siRNA and 48 hours later lysates were prepared and levels of phosphorylated Akt were analyzed by western-blot. α-Tubulin was used as a loading control; B: Densitometric analysis of P-Akt levels normalized to total Akt in control siRNA-transfected (left) or TRIP-1 siRNA-transfected cells (right) with means ± SE shown (**P < 0.05, n = 3); C: HLF-F were transfected with control siRNA or TRIP-1 siRNA, and treated right after transfection (or not) with 40 μM Akt inhibitor II for 2 days before lysates were prepared.
