A1AT pre-treatment reduces lung PA load in wild-type (WT) mice, but not in SPLUNC1 knockout (KO) mice. WT and SPLUNC1 KO mice were treated with aerosolized A1AT (0.5 mg/ml). Two hours later, mice were intranasally infected with PA (1.5 × 107 CFUs/mouse). After 24 hrs of PA infection, lung tissues were processed to quantify PA load. While A1AT significantly reduced lung PA load in WT mice (A), it did not alter the PA load in SPLUNC1 KO mice (B). N = 4 – 5 mice per group. The horizontal solid red lines indicate medians of CFUs.
